DEPARTMENT OF BIOCHEMISTRY AND MOLECULAR BIOLOGY

Beckman CEQ2000 DNA Sequencing System - Rm HSB 458. This is a high-throughput automated DNA sequencing system with 8 parallel capillary columns. It can handle up to 96 samples in a single run. The computer that runs the system is networked, so that data can be saved to MUOT servers and analyzed using various DNA sequence manipulation software programs preferred by individual users. There is no core technician, so all users are responsible for running their own sequencing reactions and loading the samples on the instruments. A per-sample fee structure has been established to cover the costs of consumable supplies (gels, buffers, capillary arrays) and the maintenance contract. The fee does not include dye-terminator kits and sequencing reagents, which must be purchased directly by the user. All users will be charged based on the number of samples run. An account number must be provided before starting any sequencing project. New users must undergo supervised training before they are allowed to operate the instrument. Time on the instrument must be reserved in advance. Additional details may be obtained by contacting Gwen Soule in Dr. Maltese's lab (x4164).

Kodak 440CF Image Station - Rm HSB 458. This is a multi-purpose digital imaging system for scanning and analyzing nucleic acid and protein gels, blots and standard x-ray films. It can detect chemiluminescence (ECL, Pierce Super Signal etc.) , fluorescence and chromogenic stains. Analysis software is located on the departmental network server (restricted access). All users are required to sign up for time in advance. New users must obtain permission and undergo supervised training. There are no consumable reagents required, but heavy users outside of the department may be asked to contribute to the maintenance contract, based on number of hours logged on the instrument. Additional information may be obtained by contacting Susan Wilson in Dr. Maltese's lab (x4164).

Centrifuges - Rms. HSB 463 and HSB 431. The department maintains a number of high-speed centrifuges and ultracentrifuges. These include two Beckman L8 ultracentrifuges, a Beckman TLX "mini" ultracentrifuge, two Sorvall RC5's, two Beckman J2-21's and a Beckman J6B. These are available for occasional use by investigators outside of the department, provided that all sign-up and log entry procedures are followed and the individual is familiar with proper use of the equipment. In all cases, users from other departments must notify one of the secretaries in the department office prior to using this equipment. Heavy users may be asked to contribute to the annual service contracts based on number of hours logged on the instrument.

Scintillation Counters - Rms HSB 431 and HSB 458. The department maintains two counters, a Beckman 3801 and a Beckman 6800.

Fluorescence Microscope and Imaging System: Rm HSB 430. The Maltese lab maintains a Nikon Eclipse fluorescence microscope with digital camera and imaging system. It is heavily used and requires supervised training. Therefore, anyone interested in using this equipment must make prior arrangements with Dr. Maltese or Dr. Overmeyer (x4164 or x4100).
